EPO Purification

Easy-to-use EPO Purification kits from MAIIA Diagnostics offer a unique system for rapid immuno-affinity purification and concentration of erythropoietins (EPO) from aqueous media such as urine, serum, plasma, dried blood spots or cellculture medium.

The anti-EPO column in the kits is immobilized with anti-EPO antibody which captures both endogenous erythropoietin (hEPO) and recombinant erythropoietin (rhEPO) such as Epoetins, NESP, CERA and EPO-Fc. The kits are intended to be used as a preanalytical sample preparation step for further analysis with techniques such as isoelectric focusing (IEF), SARCOSYL polyacrylamide gel electrophoresis (SAR-PAGE) and Membrane Assisted Isoform ImmunoAssay (MAIIA). Several purifications can simultaneously be handled if using appropriate laboratory equipment.

EPO Purification Kit (Art.1390)

25 disposable anti-EPO columns and buffers included in the kit. Suitable for few purifications per day.

Sample capacity:

10 – 20 mL urine or 0.5 – 1.0 mL serum / plasma

Purification:

Sample passes through a membrane with immobilized anti-EPO antibodies by pressure format. Flow rate 1mL / min. The bound EPO in the membrane is then released with elution buffer and spin off by using a micro centrifuge.

Purified sample:

35 – 55 µL

IEF, SAR-PAGE, MAIIA

Equipments:

0.45 μm HPF Millex HV filter and 50 mL syringe with Luer-Lok, vacuum manifold with Luer female taper connection, vacuum source and a regulator to provide a steady vacuum or similar. Microcentrifuge.

EPO Purification Gel Kit for Urine (Art.1410) or for Blood (Art.1430)

25 disposable anti-EPO gel columns and buffers included in the kit. Suitable for running many purifications simultaneously, typical 100 or more per day.

Sample capacity:

10 mL urine (Art. 1410) 
0.5 mL serum/plasma or 1-3 dried blood spot discs (Art. 1430)

Purification:

Sample incubates with anti-EPO resin by end over end incubation for 90-120 min. Aqueous media is then removed by pressure format. The bound EPO in the resin is then released with elution buffer and spin off by using a micro centrifuge.

Purified sample:

35 – 50 µL

EPO Purification Gel (Art.1271)

One regenerable high capacity anti-EPO gel column and buffers for research used. Custom quantities 2-10 mL are available.

Sample capacity:

For larger sample volumes or high EPO concentrations such as cellculture medium. Bindingcapacity 100 – 400 µg EPO / 2 mL gel depending on EPO variant.

Purification:

Sample passes through a traditional regenerable anti-EPO gel column by gravity format. Flow rate 0,5 – 3mL / min. The bound EPO is then released with elution buffer in the same way.

Purified sample:

3 – 5 mL
