EPO Purification
Easy-to-use EPO Purification kits from MAIIA Diagnostics offer a unique system for rapid immuno-affinity purification and concentration of erythropoietins (EPO) from aqueous media such as urine, serum, plasma, dried blood spots (DBS) or cellculture medium.
The anti-EPO column in the kits is immobilized with anti-EPO antibody 3F6 or 7D3 which captures both endogenous erythropoietin (hEPO) and recombinant erythropoietin (rhEPO) such as Epoetins, NESP, CERA and EPO-Fc. The kits are intended to be used as a preanalytical sample preparation step for further analysis with techniques such as isoelectric focusing (IEF), SARCOSYL polyacrylamide gel electrophoresis (SAR-PAGE) and Membrane Assisted Isoform ImmunoAssay (MAIIA). Several purifications can simultaneously be handled if using appropriate laboratory equipment.
EPO Purification Kit 3F6 (Art. 1390)
25 disposable anti-EPO columns and buffers included in the kit. Suitable for few purifications per day. The anti EPO antibody immobilized in the membrane based column is 3F6.
Sample capacity:
10–20 mL urine or 0.5–1.0 mL serum / plasma
Purification:
Buffers are added to sample and after filtration the sample mixture is transferred to a membrane based Anti EPO column with immobilized anti-EPO antibodies for pass through purification. The bound EPO in the membrane is then released with elution buffer and spin off by using a micro centrifuge.
Purified sample:
Analysis:
IEF, SDS or SAR PAGE, MAIIA
Accessories:
Equipments:
0.45 μm HPF Millex HV filter, 50 mL syringe with Luer-Lok, vacuum manifold with Luer female taper connection, vacuum source with regulator to provide a steady vacuum and microcentrifuge.

EPO Purification Gel Kit 3F6 - for Blood (Art. 1430)
EPO Purification Gel Kit 7D3 - for Urine and Blood (Art. 1460)
25 disposable anti-EPO gel columns and buffers included in the kit. Suitable for running many purifications simultaneously, typical 100 or more per day. The anti EPO antibody immobilized in the gel column is 3F6 or 7D3.
Sample capacity:
Art. 1410 3F6: 10 mL urine
Art. 1430 3F6: 0.5 mL serum/plasma or DBS samples
Art. 1460 7D3: 10 mL urine, 0.5 mL serum/plasma or DBS samples
Purification:
Buffers are added to sample and after filtration the sample mixture is transferred to anti-EPO resin for end over end incubation for 2 hours. Aqueous media is removed by pressure format and the bound EPO in the resin is then released with elution buffer and spin off by using a micro centrifuge.
Purified sample:
10-50 µL
Analysis:
SDS or SAR PAGE

Equipments:
0.45 μm HPF Millex HV filter, 20 mL syringe with Luer-Lok, vacuum manifold with Luer female taper connection, vacuum source with regulator, microcentrifuge and tube rotator for 50 mL tube.

EPO Purification Gel (Art.1271)
Sample capacity:
Purification:
Sample passes through a traditional regenerable anti-EPO gel column by gravity format. Flow rate 0.5 – 3mL / min. The bound EPO is then released with elution buffer in the same way.
